Description: IC50: 0.5 nM for Y5The actions of neuropeptide Y are mediated by at least six G-protein coupled receptors denoted as Y1, Y2, Y3, Y4, Y5, and y6. Investigations using receptor selective ligands and receptor knock-out mice show that NPY effects on feeding are mediated by both Y1 and Y5 receptors.
Description: Lymphocyte-activation gene 3 (LAG3), also known as CD223, is a type I transmembrane protein with four extracellular Ig-like domains, designated D1 to D4 and belongs to the immunoglobulin superfamily. The gene for LAG3 lies adjacent to the gene for CD4 on human chromosome 12p13.32 and shares approximately 20% identical to the CD4 gene. LAG3 is expressed on activated T cells, natural killer cells, B cells and plasmacytoid dendritic cells. LAG3 binds with high affinity to MHC class II molecules, and it interferes competitively with the binding of CD4 to MHC class II and thereby blocks the transduction of stimulatory signals mediated by this interaction. LAG3 negatively regulates cellular proliferation, activation, and homeostasis of T cells, and plays an important role in Treg suppressive function. LAG3 is the target of various drug development programs to develop new treatments for cancer and autoimmune disorders. The soluble form, sLAG-3, is being developed as a cancer drug.
Description: T-cell surface glycoprotein CD3 delta & CD3 gamma chain, also known as CD3D & CD3G or CD3D&CD3G respectively, are single-pass type I membrane proteins. CD3D, together with CD3- epsilon(CD3E) , CD3-gamma and CD3-zeta, and the T-cell receptor alpha/beta and gamma/delta heterodimers, forms the T cell receptor-CD3 complex. T cell receptor-CD3 complex plays an important role in coupling antigen recognition to several intracellular signal-transduction pathways.
Description: Natural cytotoxicity triggering receptor 3 (NCR3) is also known as Activating natural killer receptor p30, Natural killer cell p30-related protein (NK-p30), CD antigen CD337, 1C7, LY117. NCR3 /CD337 /NKp30 belongs to the natural cytotoxicity receptor (NCR) family. NCR3 /NKp30 contains one Ig-like (immunoglobulin-like) domain. NCR3 /NKp30 is selectively expressed by all resting and activated NK cells and weakly expressed in spleen. NCR3 is homodimer in the unliganted form. NCR3 interacts with CD3Z. NCR3 interacts with and is activated by binding to NCR3LG1 or BAG6. Engagement of NCR3 by BAG6 also promotes dendritic cell (DC) maturation, both through killing those DCs that did not properly acquire a mature phenotype, and inducing NK cells to release TNFA and IFNG, which promotes DC maturation.
Description: The TIM (T cell/transmembrane, immunoglobulin and mucin) family plays a critical role in regulating immune responses, including allergy, asthma, transplant tolerance, autoimmunity and the response to viral infections. The unique structure of TIM immunoglobulin variable region domains allows highly specific recognition of phosphatidylserine (PtdSer), exposed on the surface of apoptotic cells. TIM-4 (T cell; immunoglobulin; Mucin-4), also known as SMUCKLER, is a 60 kDa member of the TIM family of immune regulating proteins. TIM-4 is exclusively expressed on antigen-presenting cells, where it mediates phagocytosis of apoptotic cells and plays an important role in maintaining tolerance. TIM-4 binds specifically to TIM-1 which is also the cellular receptor for the hepatitis A virus, and has been implicated in the development of asthma. Among hematopoietic cells, TIM-1 is expressed on activated B and T cells, preferentially in the Th2 subset of CD4+ T cells. The interaction of TIM-4 with TIM-1 induces costimulatory and hyperproliferative signals in T cells.
